From Middelburg, the capital of Zeeland, it's a scenic 30-minute drive using the N57 towards Serooskerke, after which you turn onto the N287 towards Domburg via Oostkapelle. Middelburg will be on your route coming from almost all of the Netherlands, using either the A58 from Bergen op Zoom, or the more interesting N57 across the Deltawerken from Brielle and Rotterdam. From the coast of Belgium, head for Terneuzen (north of Ghent), and use the N62. This does include a €5 toll for the tunnel that crosses the Scheldt estuary, but is a significantly quicker route than the alternative journey via the ring of Antwerp.
Regular bus services run from Middelburg train station to Domburg. These are lines 52 and 53, which both operate hourly. From Ghent, an express bus route operates to this station once every two hours, and from Bruges bus line 42 connects to Breskens, where you'll have to cross the Scheldt estuary by ferry. Arriving in Vlissingen, you can take the train to Middelburg and catch connecting buses there, or use bus 56 to meet line 53 in Koudekerke.
Given its compact size, Domburg is best explored on foot or by bike.
